Using the Disposable Cell Scrapers for J774A.1 Cell Digestion Operation


J774A.1  cells are a typical type of adherent cells that are difficult to digest with trypsin. Therefore, using the disposable cell scrapers  for handling these cells will be more convenient and efficient. Here are the specific steps for using the disposable cell scrapers  to handle J774A.1 cells:


Preparation of Materials:


  • Disposable cell scrapers (unopened)


  • Clean bench


  • Centrifuge tubes, pipettes, and pipette guns (all sterilized and unopened)


Operating Procedures:


1. Pre-disinfection: Turn on the clean bench for 30 minutes for disinfection in advance, and the operators should undergo full-body disinfection according to sterile procedures.


2. Take out the materials: Take the J774A.1  cells from the incubator in a T25  culture flask, and place it with the disinfected disposable cell scrapers on the clean bench. Note that the culture flask should be placed upright and the cap should not be unscrewed in advance to prevent contamination.


3. Unpacking the scraper: Unpack the disposable cell scraper  package, hold the scraper with one hand, avoiding contact with the tabletop or other surfaces. Use the other hand to unscrew the culture flask cap, ensuring the inner side of the cap faces up to prevent contamination.


4. Scrape the cells: Insert the disposable cell scraper into the culture flask, adjust the blade direction along the cell-free wall, and gently scrape the adherent cells from outside to inside.


5. Complete the operation: After finishing the scraping, discard the cell scraper without immersing it into the culture medium. Use the pipette gun to gently blow down the cells on the flask wall and mix the cells with the culture medium, repeating 2-3 times.


6. Cell centrifugation: Transfer the cell and culture medium mixture into a centrifuge tube, then use a centrifuge for the centrifugation process. The next steps can follow the conventional passage or cryopreservation procedures.
