The anticoagulant effect of K3·EDTA is stronger than K2·EDTA. K3·EDTA results in greater RBC shrinkage and produces a larger increase in cell volume on standing.

After the tube has been filled with blood, immediately invert the tube 8-10 times to mix and ensure adequate anticoagulation of the specimen.

Anticoagulants are frequently used to stop blood from clotting in diagnostic blood tests. Because of its great ability to interact with metal ions, EDTA is frequently used to remove metal ions from solutions and modify the properties of ions. The mechanism of EDTA's anticoagulant effect is based on chelation of free Ca2+ ions, which inhibits thrombocyte aggregation and numerous hemostatic cascade steps.
EDTA has traditionally been recommended as the anticoagulant of choice for hematological assays since it preserves the most cellular components and blood cell shape.
EDTA is a calcium chelator that binds to Ca ions in the plasma to prevent blood clotting, whereas heparin inhibits the coagulation cascade by acting with a kind of thrombin.
The most frequent application of EDTA is for hematological testing (red and white blood cell count, Hb, blood group). Heparin is used in clinical chemistry assays to measure cholesterol, CRP, hormones, and other substances. If you want to perform PCR test , use EDTA since heparin interferes with PCR.
